چکیده

The 375 bp fragment of gag gene encoding fragment of matrix protein p16 of ten field isolates of maedi-visna virus (MVV) was amplified; and all the sequences were subsequently subjected to sequencing and genetic analysis. Among isolates, the homologies ranged from 87.2% to 100% and 88% to 100% at the nucleotide and amino acid level, respectively. Polish strains were more related to ovine prototypic strains with average variability of 21% in nucleotide sequence, whereas in comparison to the caprine strain CAEVCO, 33% divergence was demonstrated. Phylogenetic analysis enabled the segregation of the Polish strains into two major groups, closely related to MVVK1514 and two other ovine strains: SAOMVV and MVVEV1, confirming the presence of a complex population of MVV genotypes found in sheep from Poland.

Isolation of Maedi/Visna Virus from a Sheep in Japan

Maedi/visna (MV) is a lentiviral disease of sheep caused by the maedi/visna virus (MVV). Although MV is prevalent in many countries, it had not been reported in Japan. In 2011, however, three sheep in northern Japan were reported to be seropositive against the MVV antigen, indicating a persistent MVV infection. In the present study, we isolated MVV from one sheep to confirm MVV infection and co...

The long terminal repeat is a determinant of cell tropism of maedi-visna virus.

Maedi-visna virus (MVV) is a lentivirus of sheep, mainly affecting the lungs and the central nervous system. Long terminal repeat (LTR) sequence variability is common in tissue culture-derived isolates of MVV as well as those of other lentiviruses. The role of this sequence variation in MVV replication has not been explored. PCR amplification of the LTRs of an MVV isolate revealed two product s...

Preparation of a cell line persistently infected with maedi/visna virus and production of viral antigens

We attempted to prepare a cell line that produces maedi/visna virus (MVV) and is free of contamination by other viruses and mycoplasmas. Three cell lines, which originated from a sheep, goat and bat, were infected with MVV and passaged approximately every 5 days. The cultured cells were then subjected to polymerase chain reaction analysis for MVV provirus. As a result, a cell line persistently ...

Simultaneous mutations in CA and Vif of Maedi-Visna virus cause attenuated replication in macrophages and reduced infectivity in vivo.

Maedi-visna virus (MVV) is a lentivirus of sheep sharing several key features with the primate lentiviruses. The virus causes slowly progressive diseases, mainly in the lungs and the central nervous system of sheep. Here, we investigate the molecular basis for the differential growth phenotypes of two MVV isolates. One of the isolates, KV1772, replicates well in a number of cell lines and is hi...

Duplicated sequence motif in the long terminal repeat of maedi-visna virus extends cell tropism and is associated with neurovirulence.

Maedi-visna virus (MVV) is a lentivirus of sheep causing chronic inflammatory disease of the lungs (maedi) and the nervous system (visna). We have previously shown that a duplicated sequence in the long terminal repeat (LTR) of MVV is a determinant of cell tropism. Here, we demonstrate that deletion of a CAAAT sequence from either one of the repeats resulted in poor virus growth in sheep choroi...
